Abstract

Blood glucose testing is one of the most important laboratory examinations for the diagnosis, monitoring, and evaluation of diabetes mellitus management. The accuracy of test results is influenced by various factors, particularly those occurring during the pre-analytical phase, including the type of blood collection tube used and the delay between sample collection and analysis. Delayed examination may reduce blood glucose concentration due to ongoing glycolysis within blood cells, thereby affecting the validity of laboratory results. This study aimed to analyze the effect of delayed examination time on blood glucose levels using plain tubes and Serum Separator Tubes (SST). An experimental study with a Two-Way Repeated Measures design was conducted. Serum samples obtained from seven respondents were collected into plain tubes and SST tubes and analyzed at 0, 5, and 10 hours using a Biobase Automatic Chemistry Analyzer. Data were analyzed using the Shapiro–Wilk normality test and Two-Way Repeated Measures ANOVA. The results showed that the mean blood glucose level in plain tubes decreased from 75.57 mg/dL at 0 hours to 64.29 mg/dL at 5 hours and 65.43 mg/dL at 10 hours. In contrast, blood glucose levels in SST tubes remained relatively stable at 75.71 mg/dL, 76.86 mg/dL, and 78.14 mg/dL at 0, 5, and 10 hours, respectively. The data were normally distributed (p > 0.05). Two-Way Repeated Measures ANOVA demonstrated that examination time, tube type, and their interaction had a statistically significant effect on blood glucose levels (p < 0.05). These findings indicate that delayed examination affects blood glucose levels in plain tubes, whereas SST tubes are able to maintain blood glucose stability for up to 10 hours after sample collection.

ABSTRAK

Pemeriksaan kadar glukosa darah merupakan salah satu pemeriksaan laboratorium yang berperan penting dalam menegakkan diagnosis, memantau perkembangan penyakit, serta mengevaluasi keberhasilan terapi pada pasien diabetes mellitus. Keakuratan hasil pemeriksaan dipengaruhi oleh berbagai faktor, khususnya pada tahap pra-analitik, seperti jenis tabung yang digunakan dan lamanya waktu antara pengambilan sampel dengan proses pemeriksaan. Penundaan pemeriksaan dapat menyebabkan penurunan kadar glukosa akibat proses glikolisis yang masih berlangsung di dalam sel darah sehingga memengaruhi validitas hasil pemeriksaan. Penelitian ini bertujuan untuk menganalisis pengaruh penundaan waktu pemeriksaan terhadap kadar glukosa darah menggunakan tabung plain dan Serum Separator Tube (SST). Penelitian menggunakan desain eksperimen dengan pendekatan Two Way Repeated Measure. Sampel berupa serum dari tujuh responden yang dimasukkan ke dalam tabung plain dan SST, kemudian dianalisis pada waktu 0 jam, 5 jam, dan 10 jam menggunakan Biobase Automatic Chemistry Analyzer. Analisis data dilakukan menggunakan uji normalitas Shapiro-Wilk dan uji Two Way Repeated Measure ANOVA. Hasil penelitian menunjukkan bahwa rerata kadar glukosa pada tabung plain mengalami penurunan dari 75,57 mg/dL pada 0 jam menjadi 64,29 mg/dL pada 5 jam dan 65,43 mg/dL pada 10 jam. Sebaliknya, kadar glukosa pada tabung SST tetap relatif stabil, yaitu 75,71 mg/dL, 76,86 mg/dL, dan 78,14 mg/dL pada masing-masing waktu pengamatan. Data penelitian berdistribusi normal (p>0,05). Hasil analisis menunjukkan bahwa waktu pemeriksaan, jenis tabung, serta interaksi keduanya memberikan pengaruh yang bermakna terhadap kadar glukosa darah (p<0,05). Dengan demikian, penundaan pemeriksaan menyebabkan perubahan kadar glukosa pada tabung plain, sedangkan penggunaan tabung SST mampu mempertahankan stabilitas kadar glukosa darah hingga 10 jam setelah pengambilan sampel.
