UJI EFEKTIVITAS DAYA HAMBAT AIR PERASAN JERUK LEMON (Citrus limon L.) TERHADAP PERTUMBUHAN Trichophyton rubrum DENGAN METODE SUMURAN

ABSTRACT
Lemon (Citrus limon L.) is a fruit that contains various bioactive compounds, including flavonoids, saponins, tannins, limonene, and citric acid, which have been reported to possess antifungal properties. Trichophyton rubrum is a dermatophyte that is the primary causative agent of fungal infections affecting human skin, hair, and nails. Although the antifungal activity of lemon extracts has been widely reported, information regarding the effectiveness of fresh lemon juice against Trichophyton rubrum remains limited, highlighting the need for scientific investigation to evaluate its potential as a natural antifungal agent. This study aimed to determine the ability of fresh lemon juice (Citrus limon L.) to inhibit the growth of Trichophyton rubrum in vitro. This laboratory experimental study employed the well diffusion method on Sabouraud Dextrose Agar (SDA), with the diameter of the inhibition zone as the primary outcome variable. The results demonstrated that none of the treatments produced an inhibition zone, with a mean inhibition zone diameter of 0 mm, indicating that fresh lemon juice exhibited no antifungal activity against the growth of Trichophyton rubrum. These findings suggest that fresh lemon juice is ineffective as an antifungal agent against Trichophyton rubrum when tested using the well diffusion method. This study provides a scientific basis for future research focusing on the development of more effective extraction techniques or formulations with higher concentrations of bioactive compounds to enhance the antifungal potential of lemon.
ABSTRAK
Jeruk lemon (Citrus limon L.) merupakan salah satu buah yang mengandung senyawa bioaktif, seperti flavonoid, saponin, tanin, limonen, dan asam sitrat, yang dilaporkan memiliki aktivitas antijamur. Trichophyton rubrum merupakan dermatofita penyebab utama infeksi kulit, rambut, dan kuku pada manusia. Meskipun aktivitas antijamur ekstrak jeruk lemon telah banyak dilaporkan, informasi mengenai efektivitas air perasan segar terhadap Trichophyton rubrum masih terbatas sehingga diperlukan pengujian untuk memperoleh bukti ilmiah mengenai potensinya sebagai antijamur alami. Penelitian ini bertujuan untuk mengetahui kemampuan air perasan jeruk lemon (Citrus limon L.) dalam menghambat pertumbuhan jamur Trichophyton rubrum secara in vitro. Penelitian ini merupakan penelitian eksperimental laboratorium menggunakan metode difusi sumuran pada media Sabouraud Dextrose Agar (SDA), dengan variabel utama berupa diameter zona hambat yang terbentuk di sekitar sumuran. Hasil penelitian menunjukkan bahwa seluruh perlakuan tidak menghasilkan zona hambat dengan diameter rata-rata sebesar 0 mm, sehingga air perasan jeruk lemon tidak menunjukkan aktivitas antijamur terhadap pertumbuhan Trichophyton rubrum. Temuan ini mengindikasikan bahwa air perasan jeruk lemon dalam bentuk segar belum efektif digunakan sebagai agen antijamur terhadap Trichophyton rubrum menggunakan metode sumuran. Penelitian ini memberikan kontribusi ilmiah sebagai dasar bagi pengembangan penelitian lanjutan melalui penggunaan metode ekstraksi yang lebih optimal atau formulasi dengan konsentrasi senyawa aktif yang lebih tinggi untuk meningkatkan potensi antijamur jeruk lemon.
